PQQ: The Compound That Grows New Mitochondria

Pyrroloquinoline quinone (PQQ) occupies a distinct niche among mitochondrial-support supplements. While compounds like CoQ10 support the function of existing mitochondria, PQQ has a separate and complementary mechanism: it appears to stimulate mitochondrial biogenesis — the process by which cells create entirely new mitochondria.

What Is PQQ?

PQQ is a redox cofactor found in trace amounts in fermented soybeans (natto), parsley, green tea, kiwi fruit, and human breast milk. It was originally studied as a nutrient in bacteria before its relevance to mammalian and human biology became clear. Unlike vitamins, PQQ has not been officially classified as an essential nutrient, but a growing body of evidence suggests it plays meaningful physiological roles, particularly in mitochondrial health.

The Mitochondrial Biogenesis Mechanism

Mitochondrial biogenesis is regulated substantially through a signaling pathway involving PGC-1alpha, a transcriptional coactivator often described as the master regulator of mitochondrial production. Animal studies have found that PQQ supplementation activates this PGC-1alpha pathway, leading to measurable increases in mitochondrial density in tissues including the liver and brain [1].

This is mechanistically distinct from — and complementary to — how compounds like CoQ10 work. CoQ10 supports the electron transport chain within existing mitochondria; PQQ appears to influence how many mitochondria a cell has in the first place. The distinction matters because mitochondrial density naturally declines with age, and a cell with more (healthy) mitochondria has more total capacity for energy production, independent of how efficiently each individual mitochondrion operates.

Animal Evidence

The foundational research establishing PQQ's mitochondrial effects came from studies feeding mice PQQ-deficient diets, which resulted in significantly reduced mitochondrial content in tissue compared to mice fed PQQ-adequate diets — the deficient mice showed impaired growth, reproductive performance, and immune function that improved substantially when PQQ was reintroduced to the diet [2].

Subsequent studies specifically supplementing PQQ (rather than just avoiding deficiency) found increased mitochondrial biogenesis markers, improved mitochondrial function, and protection against oxidative damage to mitochondrial DNA in various animal models [1].

Cognitive and Neuroprotective Research

Given the brain's exceptionally high energy demands and dependence on mitochondrial function, PQQ has been studied specifically for cognitive effects. A randomized controlled trial in middle-aged and older adults found that PQQ supplementation (20mg/day) improved measures of working memory compared to placebo over 12 weeks, with additional benefit observed when PQQ was combined with CoQ10 compared to PQQ alone [3].

Animal studies have found PQQ protects neurons against oxidative stress-induced damage and may support the survival of neurons under conditions that would otherwise trigger cell death, though translating these neuroprotective mechanisms into confirmed human clinical outcomes for conditions like cognitive decline requires further large-scale trials.

Antioxidant Properties

Beyond its mitochondrial biogenesis role, PQQ is itself a potent antioxidant with an unusual property: unlike most antioxidants, which are consumed in the process of neutralizing free radicals and require replenishment, PQQ can undergo thousands of redox cycles without being degraded, making it exceptionally efficient on a per-molecule basis in laboratory studies [4].

PQQ and CoQ10: A Common Pairing

Given their complementary mechanisms — PQQ potentially increasing mitochondrial quantity, CoQ10 supporting the function of each individual mitochondrion — the two are frequently paired in longevity-focused supplementation. The clinical trial noted above found enhanced cognitive benefit when PQQ was combined with CoQ10 relative to PQQ alone, offering some direct evidence supporting this combination approach rather than it being purely mechanistic speculation [3].

Sleep and Mood Research

Some research has also explored PQQ's effects on sleep quality and mood, with a small trial finding improvements in sleep quality and reduced markers of mental and physical fatigue in adults supplementing with PQQ over 8 weeks [5]. This research area is less developed than the mitochondrial and cognitive research, and larger trials would help confirm these preliminary findings.

Dosing

Clinical trials have generally used 10-20mg/day of PQQ, typically as PQQ disodium salt (the most common commercial form, sometimes branded as BioPQQ). Effects on mitochondrial biogenesis are thought to require sustained supplementation given the gradual nature of building new mitochondrial mass, rather than producing acute effects from a single dose.
